Dermal Delivery of Niacinamide—In Vivo Studies

In vivo human studies are considered to be the “gold standard” when investigating (trans)dermal delivery of actives. Previously, we reported effects a range vehicles on niacinamide (NIA) using conventional Franz cell studies. present work, dermal NIA was investigated in subjects confocal Raman spectroscopy (CRS) and tape stripping (TS). The included propylene glycol (PG), Transcutol® P (TC), binary combinations PG with oleic acid (OA) or linolenic (LA) ternary system comprising TC, caprylic/capric triglyceride (CCT) dimethyl isosorbide (DMI). For CRS studies, higher area under curve (AUC) values for were observed PG:LA compared PG, TC TC:CCT:DMI (p < 0.05). A very good correlation found between vitro cumulative permeation AUC from intensity depth profiles, Pearson coefficient (R2) 0.84. addition, an excellent (R2 = 0.97) evident signal solvent active. also shown discriminate solution versus crystalline NIA. findings confirm that is emerging as powerful approach dermatopharmacokinetic both actives excipients human.
